Introduction
SLS' gDNA Extraction kit(mini) provide user friendly and easy methods for purification of total DNA from small amount of samples for various downstream processing.
This simple protocol which is ideal for simultaneous processing of multiple samples, yield pure DNA ready for direct amplification in few minutes. This protocol requires no phenol/chloroform extraction or alcohol precipitation, and involves very little handling. DNA is eluted in Buffer DE, ready for direct addition to PCR or other enzymatic reactions. Alternatively, it can be safely stored at –20°C for later use.
The purified DNA is free of protein, nucleases, and other contaminants or inhibitors.
Principle
SLS’ gDNA Extraction kit(mini) is designed for rapid purification of total DNA (e.g., genomic, mitochondrial) from small quantity of samples.
Cells are lysed during a short incubation with Proteinase K in the presence of a chaotropic salt, which immediately inactivates all nucleases. Cellular nucleic acids bind selectively to glass fibre membrane in a special centrifuge tube. The nucleic acids remains bound while a series of rapid “wash-and-spin” steps which removes contaminating small molecules. Finally, low salt elution removes the nucleic acids from the glass fibre membrane.
The process does not require precipitation, organic solvent extractions, or extensive handling of the nucleic acids.

Storage Conditions and Product Stability
The Proteinase K should be stored at -20°C upon arrival or after reconstitution.
All other solutions should be kept tightly sealed and stored at room temperature. These reagents should remain stable for at least 1 year in their unopened containers.
